چکیده

The production of hydrogen ions during the hydrolysis of ATP by myosin B and the consequent fall in pH can produce alterations in enzymatic activity directly, or by influencing Ca ion concentrations controlled by metal-chelate systems. Adequate hydrogen ion buffer systems can limit pH changes to acceptable levels. However, several commonly used hydrogen ion buffers have been reported to have specific effects on contractile protein responses, and all buffers can indirectly change activity through their contributions to the ionic strength. In this study, ten potentially useful buffers suitable for pH control between pH 5 and 9 were evaluated for their effects on myosin B ATPase activity. In contrast to previous reports, all were found to have no effect on this system if both the buffering value (AH+/apH) and the ionic strength were maintained constant. Simple methods are presented for determining the total and ionized concentrations of any buffet pH control at any operating pH.

The Inhibition of the Adenosine Triphosphatase Activity of Actomyosin by Magnesium Ions

In 1939 Engelhardt and Ljubimova (2) suggested that myosin (the contractile substance of muscle) and adenosine triphosphatase are probably identical, and that 'the calcium ion is a specific activator of this enzyme. Working with a myosin suspension made from rabbit muscle (by the method of Greenstein and Edsall (4) or of Weber and Meyer (11)) D. M. Needham (9) tested the activating powers of bo...

Cardiac myosin adenosinetriphosphatase of rat and mouse. Distinctive enzymatic properties compared with rabbit and dog cardiac myosin.

Cardiac myosin obtained from rats and mice (smaller animals) had a higher adenosinetriphosphatase (ATPase) activity in the presence of calcium ions (Ca) than did cardiac myosin from rabbits and dogs (larger animals). Structural differences between the two types of cardiac myosin were suggested by the lower apparent activation energy of the Caactivated ATPase reaction catalyzed by cardiac myosin...

The Influence of Ethylene Glycol on the Enzymatic Adenosine Triphosphatase Activity and Molecular Conformation of Fibrous Muscle Proteins.

Considerable interest has recently been evoked in the study of proteins in nonaqueous solvents, very largely as a result of the observations of Rees and Singer (1) and Yang and Doty (2) that the internal configuration of protein molecules could be radically altered in nonaqueous media, as reflected by changes in sedimentation constants, viscosity increments, and optical rotatory dispersion. Of ...

Effect of the thyroid state on the enzymatic characteristics of cardiac myosin. A difference in behavior of rat and rabbit cardiac myosin.

The effect of thyroid state on the activity of myosin adenosinetriphosphatase (ATPase) was examined in the rat and the rabbit. Cardiac myosin from thyroxine-treated rabbits showed enzymatic properties characterized by high Ca2plus-activated ATPase activity, low activation energy, lower rate of inactivation at alkaline pH, and no activation by N-ethylmaleimide compared with the same properties i...
